The Coagulase Test
- Possession of the enzyme coagulasewhich coagulase plasma is an almostexclusive property of Staph.aureus.Thereare two ways of performing this test–The Slide Coagulase Test–The Tube coagulase Test
第13页:The Coagulase Test
The Coagulase Test
- Coagulase is an enzyme convertingfibrinogen into fibrin promoting bloodclotting. It has been speculated thatthis enzyme might be a virulence factorwith the coagulated blood around thebacteria protecting them from the immunesystem.
- However, coagulase-negative strains areoften as pathogenic as coagulase-positive strains.

第16页:The Dnase Test
The Dnase Test
- Inoculate Dnase agar plates with a loop so thatthe growth is in plaques about 1 cm indiameter.Incubate at 370C overnight.Flood theplate with 1 N hydrochloric acid.Clearingaround the colonies indicates Dnaseactivity. The hydrochloric acid reacts withunchanged deoxyribonucleic acid to give acloudy precipitate.A few other bacteria,e.g.Serratia,may give a positive reaction.
第17页:The mannitol fermentation
The mannitol fermentation
- Inoculate the bacteria into a mannitolmicro-tube,incubate at 370C for18h.S.aureus will ferment mannitol toproduce acid, which causes the medium toturn yellow.
